N/A

Tbr2 Anitbody was used to specifically mark intermediate progenitors of a developing neocortex at E12.5 stage of embryonic development.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Frozen embryonic mouse brain tissue (E12)

Primary Incubation

1:400, 4 degrees overnight

Blocking Agent

10% Normal donkey serum in PBST (0.5% Triton X 100)

Secondary Incubation

1:500, AF 488

Tertiary Incubation

None

Detection

Immunofluorescence

Results Summary

Basal/Intermediate progenitors from the E12.5 developing mouse cortex were specifically labelled with this antibody. Antigen retrieval using Na.Citrate buffer (samples cooked in microwave with the buffer-2-3 min boiling) improves the staining significantly. Free floating sections could be incubated for longer time in blocking buffer (with 0.5% Tritonx-100).
