Clone Number:

N/A

E13 embryonic mouse brain tissue was stained with anti-PCNA to determine the total proliferating cells in the cortex.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Frozen embryonic mouse brain tissue (E13)

Primary Incubation

1:800, 4 degrees overnight

Blocking Agent

10% Normal donkey serum in PBST (0.5% Triton X 100)

Secondary Incubation

1:500, AF 488

Tertiary Incubation

None

Detection

Immunofluorescence

Results Summary

Proliferating cells were specifically marked with the antibody. A few cells show punctate staining with the antibody. these are the cells in the S-Phase of the cycle, hence one can also determine the total S-phase cellular population in a specific tissue.

Additional Notes

Antigen retrieval in 10mM Sodium citrate buffer (pH 6) is required for improved staining.
