E11 Frozen embryonic mouse brain sections were blocked with (10% Normal donkey serum in PBST (0.5% TritonX 100)). Primary antibody (1:1000) was used in the same buffer as blocking and the sections were incubated at 4 degrees O/N. fluorescently conjugated AF488 secondary was added after 3 washes of PBS (10 min each) and incubated for 2h at RT. Slides were washed and mounted using fluormount.The antibody does not work well with thick vibratome sections upto 250 µm as the penetration of the antibody is not good.

The antibody is highly specific and works in 20µm thin sections. radial gial fibers are visible in the image that run parallel from apical to basal area of the neocortex.
