Product Name:

NAD/NADH Assay kit (colorimetric)

Catalog Number:

ab65348

This NAD/NADH assay kit supports material like urine, serum, or lysates, and most importantly gives a quantitative NAD/NADH ratio. I use it for testing the NAD/NADH ratio in tissues and serum, which requires precise purification. It is relatively convenient and easy to optimize, with the capability to quantify NAD and NADH individually or their ratio, and have no background counting from NADP or NADPH.The best part of this kit is if your sample will have autofluorescence, it does not read it. The drawback is the sensitivity of this method since a colorimetric assay was used, it is not as sensitive as a fluorometric assay.

Experimental Design and Results Summary

Application

Ratio in mouse tissue and serum.

Starting Material

Mouse with different treatment.

Tips

Do your calculations for your standard curve ahead of time; flash freeze your samples if cannot be done in the same day.

Results Summary

I want to see if there is any difference in NAD/NADH between control and treated mice, I collected tissue and serum from mice, flash freeze them, and then the second day I processed them with this kit. The result shows a relatively good number comparing to several publications.

Summary

The Good

No fluorescent interference; detailed protocol provided in the kit.

The Bad

Expensive, procedure is very long.

The Bottom Line

NAD/NADH ratio is hard to measure, this kit gives a good result for tissue and serum.
