Normal Goat Serum

Normal goat serum was used to prepare blocking buffer in immunohistochemistry. For preparing blocking buffer, normal goat serum was prepared in 3% BSA in 1:20. At first tissue sections on the slide were treated with triton X-100, and the followed by blocking with blocking buffer. Thereafter, it is incubated in primary antibody, then secondary antibody and finally stained with DAPI. After air drying the slides, the tissue section is covered by mounting medium and covered with coverslip.

One hour incubation with blocking buffer was sufficient for proper blocking
Slides were observed under the microscope and the staining done was specific with no background signal.
