Human tumor specimens were dissociated with the Miltenyi Tumor Dissociation it. As there were concerns about sticky DNA adhering to viable cells and clogging the strainer, DNase I was employed to remove it.

Make sure to carefully calculate the amount needed as the quantification is measured in units (or Kunitz units). PBS for dissolving DNase I works fine at 5 mg/mL.

Results Summary

No viscosity was apparent and the cell strainer did not get clogged when run the dissociated tumor cells through.

DNase I from bovine pancreas is an effective enzyme for digesting DNA to help clear it and prevent obstruction of downstream analyses. It can be easily dissolved in PBS (however, this may not be stored for a very long time as enzyme activity may decrease dramatically after a few months in -20 degrees).
