The aim of the experiment was to detect protein concentration in supernatant of cell lysate. BCA reagent was prepared by mixing reagent A, B and C according to the manufacturer’s instruction. Different volumes of bovine serum albumin (2 mg/ml) as a standard and 5 microlitre of sample were taken in glass tubes. Then, 1 ml of BCA reagent was added to all the tubes, covered with foil and incubated at 60°C for 1 hour in water bath. Absorbance were read at 562 nm.

Standard/sample and BCA reagent should be mixed thoroughly. After adding BCA reagent, all the tubes were covered with aluminium foil.
The absorbance were taken at 562nm. From the standard graph, the concentration of protein in sample is calculated. The readings taken in replicates showed no deviation, results were reproducible

Sensitivity of the assay is 0.5 µg/mL and its easy to perform the experiment.
Excellent kit for estimating protein concentration with good sensitivity.
