Company:

Thermo Fisher Scientific

Product Name:

CD34 Monoclonal Antibody (4H11), PE-Cyanine7

Catalog Number:

25-0349-41

Clone Number:

4H11

CD34 is expressed on human hematopoietic stem cells, which are crucial for the generation of bone marrow elements. In our research we identify these cells to look at their expression of other markers. We need a good antibody conjugated to a fluorochrome that is not one of the common ones (FITC, PE, APC), so we got this PE-Cy7 antibody.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Human bone marrow specimen

Primary Incubation

5uL antibody at 4°C for 15 min, followed by wash and lyse

Blocking Agent

None

Secondary Incubation

None

Tertiary Incubation

None

Detection

Flow Cytometry (PE-Cy7 channel = 780/60)

Results Summary

The results were acceptable. At 5uL the separation of stem cells from non-stem cells was OK. Even at 2.5uL, the resolution was seen, but the two peaks were too close to each other, which is why we selected 5uL. It did lead to some spread into the PE channel, which was expected, but this made the resolution in that channel harder.

Summary

The Good

Bright signal.

The Bad

Lots of spillover/spread into PE channel

The Bottom Line

The antibody worked, and we will be using it. There might be better options out there!
