The NEXTflex DNA barcodes can be used with the NEXTflex Rapid DNA-Seq Kit for the generation of genomic DNA libraries that can be used in multiplexed Illumina sequencing applications. The barcodes can be used with and without additional PCR amplification cycles to accommodate samples with both low (1 ng) and high (1 ug) concentrations of starting material.

While this barcode kit has been used by our group with the NEXTflex Rapid DNA-Seq Kit 2.0, it is validated for use with other Bioo Scientific library preparation kits as well.

Results Summary

Successful library preparation can be determined by qPCR (e.g. quantity of adapter ligated library fragments generated) and TapeStation (e.g. size and distribution of library fragments). In general, the results generated from this workflow are consistently high for users in our group.

The Good

Kits are available with variant numbers of unique barcode adapters (e.g. 6, 12, 24, 48, etc.); All vials contain enough reagent for up to 8 reactions; The barcodes can be used with or without additional PCR amplification cycles and are therefore amenable to a wide range of sample concentrations.

The Bad

The barcodes are expensive.

The Bottom Line

We endorse the use of the NEXTflex DNA barcodes in the NEXTflex Rapid DNA-Seq library preparation protocol. We have had success with sequencing samples on the low end of the input range (e.g. 1 ng) and are pleased that this kit gives us the ability to multiplex our Illumina sequencing runs via a simple and intuitive workflow.
