Product Name:

Human Apoptosis Signaling Array C1

Catalog Number:

AAH-APOSIG-1-2

This apoptosis kit was very user-friendly and gave detailed instructions on how to analyze findings using a software like Excel. This assay was very helpful for examining multiple gene targets simultaneously, so we got a lot of data out of 1 experiment. Negative and positive controls were included on the blots, which was useful in subsequent analyses. This assay gives you a lot of data regarding the levels of important proteins involved in cell survival and cell death signaling pathways. For interpretation of results, it would be helpful if the manufacturer denoted which proteins specifically are involved in apoptosis and which may be associated with inhibiting apoptosis. Otherwise, I found this to be a very easy to use kit with clear instructions and I would recommend this for discovery purposes. Notes: the Caspase 3 antibody seemed a lot more sensitive than other antibodies on the blot. Also, some of the gene names in the kit manual differed from those listed on the website, so keep that in mind when analyzing data to ensure that the correct gene is being referred to.

Experimental Design and Results Summary

Application

Discovery assays, cell death, cell survival, cell signaling

Starting Material

Cell lysates

Tips

Mean dot intensity data can be hard to interpret, so I would suggest normalizing data to a proper control.

Results Summary

Our results showed several pro-apoptotic proteins were altered in our cell system.

Summary

The Good

Ease of use, requires minimal protein input, so it's good for smaller samples.

The Bad

The cost prohibits the use of replicates, therefore I had to pool biological replicates on a single blot, so my data does not have statistical significance.

The Bottom Line

I would recommend this product to others looking to assess a panel of apoptosis signaling proteins at one time.
