The transformed E. coli cells were expended to 250 ml and centrifuged to obtain cell pellets. These pellets were used to extract plasmid DNA using PureLink™ Fast Low-Endotoxin Maxi Plasmid Purification Kit as per the manufacturer's instructions. Whole steps were followed with precision and isolated plasmid DNA was checked for its quality and quantity.

The isolated plasmid DNA was checked for qualitative and quantitative parameters. NanoDrop analysis has revealed that even the purity of DNA was high the quantity was extremely low, and the values were almost 40ng DNA per microliter. This DNA came from 250 ml of culture media.
