Tissue treated with an inhibitor was lysed in QIAzol reagent and total RNA was extracted as per manufacture instructions. In brief, the tissue was first washed in cold PBS and homogenized in QIAzol buffer. To the tissue homogenate, chloroform was added, mixed well, incubated and RNA was precipitated with chilled isopropanol. The resulting mixture was centrifuged, and the pellet was resuspended in nuclease-free water for further downstream processing.

The reagent has extracted good quality RNA from cells with a 260/280 ratio of 2.3 and a 260/230 ratio of 2.1. Almost 160 micrograms of RNA were extracted from 10 million cells.
