Protein extracted from control and treated samples were resolved in 10% gel and immunoblotted with the specific antibody. Immunoblot was probed with an HRP-conjugated secondary antibody and washed. Amersham ECL Western Blotting Detection Reagent 1 and Reagent 2 were mixed in a ratio of 1:1 and the membrane was dipped in this solution for 1 minute. The luciferase signal was captured in an x-ray film.

Develop the immunoblot membrane for 1 min at room temperature.
Got a very good signal from the bands where HRP-conjugated secondary IgG bound with its target. The signal lasts for at least 1hour from the time of the development.
