To study the gene expression levels in sorted CD4+ T cells from splenocytes of vaccine exposed and control mice, we have used this cDNA synthesis kit. Total RNA was isolated, quantified and checked for the RNA purity. To prepare the cDNA, 1 µg of total RNA was used from each sample in 20 µl reaction mixture and run in thermo-cycler at 37ᵒC for 1 h. This cDNA was used to perform the quantitative PCR of different targeted genes in both the groups.
