The mouse tissues were homogenized, sonicated, and proteins were extracted in RIPA buffer. The extracted proteins were measured, and an equal amount of proteins were resolved in 7% SDS-PAGE. High range protein marker was used for this experiment. Resolved proteins were further immunoblotted for the detection of ACCβ protein using specific antibody obtained from Santa Cruz Biotechnology. The selection of an antibody was based on the merits shown in antibody flyer.

Although antibody had detected a few off-target non-specific epitopes, the specificity was very high for its on-target ACCβ.
