PCR is a technique widely used in molecular biology, microbiology, genetics, diagnostics, and many other applications. It’s very important to use appropriate reagents and instruments to get reliable results. The QB96 fulfills this role thoroughly. This is one of the most versatile and affordable thermal cyclers available. We are having been using this instrument for almost 16 months without any problems.

We have the QB96 gradient thermal cycler with satellite option. The QB96 comes with interchangeable thermal blocks (optional): 96-well, 0.2 ml tubes or microplate; 48-well, 0.5 ml and 48-well, 0.2 ml (combi); 384-well microplate and microarray/slide. We use the 96-well 0.2 ml blocks. Amongst various unique features of the QB 96, the one I like most is the lid which can be adjusted manually on the basis of the height of the tubes. This definitely prevents evaporation (which is an important factor during PCR). The QB96 is operated via an external PC through a USB network; if required, 4 thermal cyclers can be controlled from a single PC. This is done simply by connecting USB keys to the USB ports at the back of the instrument.

QBPC software is provided with the instrument. The software has a unique cycle wizard which enables stepwise protocol definition, with no tedious calculations required. Drag-and-drop icon-based commands make all steps quick, easy, and intuitive throughout. A gradient protocol can be set up with only a single point and click.

The software comes with the following distinctive features:
Automatic/manual hot start which enables the performance of an extended hot start without degrading the enzyme.
Touchdown PCR one-step command to reduce non-specific binding.
Gradient can be set up across a temperature range of up to 30°C
Program Wizard for generating your own PCR program on the basis of primer Tm. All we need to provide following inputs: biological data for the primer and template DNA (such as primer sequence forward and reverse), origin of DNA (eukaryote/prokaryote) and type of Taq (normal/hot start). The software automatically generates entire PCR profile using melting temperature calculations. This Program Wizard is an ideal tool for first shot PCRs.

The maximum heating and cooling rates are 5°C per second and 3.5°C per second which provide faster turnaround times. We use this instrument for routine PCR, including protocol optimization for multiplex PCR, RT-PCR and sometimes for cloning. We generally use 0.2 ml tubes with a 20 ul reaction volume.

I find the QB96 is an ideal instrument for any PCR-related work. It can fulfill all requirements from routine PCR to protocol optimization.
