The GeneBall™ Genome Preparation Kit is designed to extract highly purified DNA from blood or cultured cells in a quick and simple manner. By utilizing a special bead, the GeneBall™, this kit achieves highly efficient and reproducible recovery of DNA with high purity and little damage to the DNA. The GeneBall™ is made from silica and binds DNA in the presence of certain salts and under certain pH conditions. One GeneBall™ is used for each purification. As the bead diameter is 6.5 mm, the bead can be separated from liquid by gravity. Accordingly, washing is quite simple. The best feature of this kit is there is no centrifugation for washes or final elution.
Anticoagulants such as EDTA, citric acid, heparin are compatible with the kit. The steps involved in the extraction are very simple: Add Solution 1 to 8 ml of blood diluted in PBS, centrifuge and discard the supernatant. Add one GeneBall™, followed by Solutions 2 and 3, vortex and incubate at 65ºC for 20 min (in a dry block on the lab bench). Next, wash with ethanol; make sure to wait for approximately 1 min after the addition of ethanol to let the beads settle. Repeat the wash 2 to 3 times. Discard the supernatant and add TE buffer, vortex and incubate for a few minutes. The supernatant contains the DNA.
The entire procedure is quick and simple; the best part about the kit is the entire procedure can be completed in a single tube. Some components of this kit have to be stored at -20ºC. Care must be taken when pipetting so that the tip of the pipette does not touch the bead; this might lead to a loss of DNA. We were able to extract 30 – 60 μg from 1 ml of whole blood with this kit. The purified DNA generally gives an A260/A280 of 1.8 - 2.1.
We have started using this kit for extraction of genomic DNA from whole blood. This DNA is then used for molecular typing by PCR. The yield of DNA obtained from this kit is very high. As centrifugation is not required for washing, a lot of time is saved. This kit has given us good, reproducible results. I would recommend this kit to all the labs who want to save time and have good workload.
