I have used silicone for sealing the coverslip and slide. Just make sure to used the type I silicone, others interfere with prolong reagent and change the colour of mounting media

Slides containing the cells in 3D cultures were IF stained and mounted with Prolong gold antifade reagent. It mounts the samples well and protects the fluorophores from light.

Prolong gold antifade is a good mounting media, which protects the sample for longer duration. It protects the fluorophores used from bleaching when exposed to lasers for a long duration.

It is expensive

I recommend using Prolong gold antifade reagent for mounting because of its efficiency. Even though it's expensive, the samples can be imaged even after a month when using this product with good fluorescence.
