Antibodies are produced by plasma cells (PC) that are terminally differentiated B cells and play crucial roles in host protection against infections but are considered to mediate autoimmunity in patients with rheumatic diseases, such as systemic lupus (SLE) or rheumatoid arthritis (RA). Our research aims at understanding the biology of antibody-secreting cells (ASC) and B cells in health and disease. Here, we aimed at staining immature and mature B cells.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

C57BL/6 spleen and BM

Primary Incubation

Surface staining with CD93 PE-Cy7 (1/200), 20 min at 4C

Blocking Agent

Results Summary

As the majority of the B cells in the BM are immature they stain positive for CD93, with only a fraction of recirculating mature B cells being negative for CD93. In the spleen, the great majority of the B cells are mature and negative for CD93.
