To study the cell cycle, we have stained the control and treated human neuroblastoma cell lines with propidium iodide dye. After completion of toxin treatment cells were trypsinized, washed twice with PBS and fixed in 70% ice cold ethanol for 24 h at -20 degree C. After completion of cell fixation in ethanol, cells were washed twice with PBS and incubated with PI staining buffer containing propidium iodide, RNAse, 0.05% tritonX-100 in PBS for 30 minutes in dark. After that cells were analysed by flow cytometer in FL3 channel.
