We study carcinogenesis in the PyMT-driven mammary gland tumor mouse model. To visualize exhausted lymphocytes in tumors, we tested this polyclonal goat a-PD1 antibody in tumors, with excellent results.
Immunofluorescence
PLP fixed mouse (C57BL/6) PyMT tumor cryosections
1 in 50 for 3 hr at room temperature
1% BSA, 1% FCS, 0.3% Triton X, 2% mouse serum, 2% rat serum, 2% donkey serum
Donkey anti-goat Alexa Fluor 647 (1:500) for 1 hr at room temperature
Images were acquired using an inverted Zeiss LSM880 confocal microscope with an 20x 0.8NA air objective.
Good staining of exhausted T cells: CD3 (green), PD1 (red), Hoechst (blue)
