Company:

Thermo Fisher Scientific

Product Name:

Aggrecan (PG) Human ELISA Kit

Catalog Number:

KAP1461

I have used this kit to assess expression of Aggrecan protein in cell culture supernatants taken from human articular chondrocytes cultured with varying conditions. The test is sensitive to protein produced by 1x10^5 cells cultured for 5 days and generates high confidence results with a consistent standard curve.

Experimental Design and Results Summary

Application

Assessing aggrecan expression in cell culture supernatants

Starting Material

Human articular chondrocytes

Tips

The whole ELISA plate does not need to be used in one application. Enough standard is supplied to use half the plate in one assay.

Results Summary

Positive controls supplied with the kit give additional confidence and fell within range. The test is sensitive to protein produced by 1x10^5 cells cultured for 5 days and I generated results from all test samples.

Summary

The Good

Sensitive assay for detection of aggrecan in cell culture supernatants

The Bad

The sample dilution requires initial optimisation. The kit is time consuming to use for the first time, considering prep time to dilute all the reagents.

The Bottom Line

A simple, sensitive and effective kit for assessing aggrecan expression in cell culture supernatants. The supplied positive controls give additional confidence.
