Thermo Scientific™ Shandon™ Kwik-Diff™ Stain

I have used the Thermo Scientific™ Shandon™ Kwik-Diff™ Stain to stain chondrocytes in order to assess migration across a membrane in response to stimulus. My method involved fixing the cells using ice cold methanol for 5 x 1s. These were then lowered the into Diff-Quick Stain 1 for 5 x 1s. gently blotted, lowered into Diff-Quick Stain 2 for 5 x 1s, blotted, lowered into Diff-Quick Stain 3 for 5 x 1s, blotted, and thoroughly rinsed in PBS. The cells were then dehydrated with gradient ethanol washes before drying and assessing.

Careful blotting should be performed to prevent cross contamination of dyes.
The cells were clearly stained and detectable directly from microscopy of the membrane, without need for slide mounting.
