Elevated circulatory nitric oxide levels play detrimental role in sepsis. Therefore, we aimed to evaluate role of macrophages in inducible nitric oxide synthase (iNOS) expression and further to inhibit it with 1400W.

RAW macrophages were treated with 200 ng/ml LPS alone or after pre-treatment with 1 μM 1400W (30 min pre-incubation) for 8 h to follow iNOS expression using Western blot.
Worked as expected.
LPS treatment elevated iNOS expression while 1400W pre-treatment significantly blunted this effect.
