My research focuses on molecular mechanisms of axon growth and guidance during neural development and axon regeneration after injuries to the central nervous system or peripheral nerves. For this experiment, I want to detect the expression of myosin IIB in retina ganglion cells.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Mouse retina cryostat sections

Primary Incubation

1:100, overnight at 4°C

Blocking Agent

PBST (0.3%) containing 10% goat serum

Secondary Incubation

1:500, 1 hr at room temperature

Tertiary Incubation

N/A

Detection

CCD camera connected to a Zeiss inverted fluorescence microscope controlled by AxioVision software

Results Summary

I compared the signal of myosin IIB (green) in control group and in myosin IIB knock-out group. Under the same exposure time, the signal of myosin IIB in myosin IIB knock-out group is rather weaker than that in control group. However, there is still lots of unspecific binding. Moreover, this antibody can not be used to detect myosin IIB by western blot.

Summary

The Good

Myosin IIB can be detected by immunofluorescence staning

The Bad

Non-specific

The Bottom Line

If you can find an antibody that is more specific, I wouldn't recommend this one.
