Company:

Promega

Product Name:

Wizard® SV Gel and PCR Clean-Up System

Catalog Number:

A9282

Main goal is to purify the specific DNA product from gel after doing PCR or restriction digestion reaction and use it further for ligation purposes.

Experimental Design and Results Summary

Application

Extracting size-specific DNA bands from gel

Starting Material

1 microgram of DNA

Tips

To increase the yield: incubate the the elution buffer/water before the last step at 50-60c and use it for loading into the column.

Results Summary

The selected DNA bands from gel are cut and extracted using this kit. The eluted product concentrations are good (~50-100ng/ul) when the starting DNA used was ~1ug. When used less than 1ug of DNA the final eluted product concentrations were very low (less than 20ng/ul).

Summary

The Good

Easy steps and can be done within 15mins

The Bad

Poor yield after gel extraction : when used less than 1 microgram of DNA

The Bottom Line

Overall good for gel extraction and excellent for PCR product purification. I recommend this kit if you are doing cloning on daily/weekly basis.
