My main goal is to create mutations in DNA sequence finally to alter the amino acid sequence of the protein. I used this kit many times to create a single mutation in the DNA sequence, and it worked perfectly almost every time with high efficiency of mutation. I also used the same kit to create multiple site-directed mutations (single primer with multiple mutations) and it worked fine.

Tips

I used only 1 micro liter of DpnI enzyme instead of two and increased incubation time - works perfectly.

Results Summary

I did almost 20 reactions to create different single and multiple mutations in plasmid DNA (plamsid sizes: 6-12kb) and all of them worked perfectly with no extra unwanted mutations. Heat shock method of transformation with other E.coli strains other than the provided XL-gold strains also worked perfectly for this kit.
