I am trying to perform an experiment to check/verify the binding of cyclic-di-GMP with our specific protein of interest. For that purpose I used this biotinylated c-di-GMP to perform a pull-down assay using streptavidin beads and tested the eluted product by running SDS-PAGE.

Experimental Design and Results Summary

Application

Biotin-avidin based protein pull down assays/Immunoassays

Starting Material

Purified protein (His-tagged) from Bl21De3 E.coli strain

Tips

Always store in freezer after dilutions

Results Summary

Used as low as 100 pico moles concentration and still the band intensity is very high and showed excellent affinity with streptavidin beads. I suggest starting with very low concentrations (even less than 100 pico moles) for pull down assays with strepatavidin.

Summary

The Good

Solubility and concentration provided

The Bad

Product stability at room temperature

The Bottom Line

Good product with good solubility and with high percentage of purity. Even though it says that product is stable at room temperature, stability is kind of an issue if stored outside for longer periods.
