Parvalbumin interneurons regulate excitatory information processing in the brain. High expression of parvalbumin in these neurons indicate robust activation and regulation shifting the E/I balance. The antibody was used to determine developmental differences at different mouse ages. It is an easy antibody to use with a good dilution of 1:1000.

Primary Incubation

Slices were then quenched with 50 mM ammonium chloride for 15 minutes and washed with PBS

Blocking Agent

Nonspecific staining was blocked with a 5% NGS and 1% BSA in 0.1M PBS solution

Secondary Incubation

Incubated in antibody for 24 hrs at 4 degrees in 0.1 M PBS containing 1% NGS, 0.5% BSA, and 0.1% Tween-20 solution

Tertiary Incubation

Donkey anti-mouse Alexa with a dilution of 4µg/ml in 0.1M PBS for 1 hr at room temperature

Results Summary

The antibody is able to stain Parvalbumin interneurons in brains of P14 and higher aged mice. Stand alone counts and colocalization counts were easy to do with a confocal microscope at 60x.

I would recommend this antibody as the quality is good with no nonspecific binding. A decent dilution where too much antibody is not required for detection of PV cell is a bonus.
