Our lab is interested in studying the role of inflammatory kinases as a therapeutic target. We perform ELISA and western blot assays to determine the role and pro-inflammatory status of macrophages in response to pharmacological inhibition of these kinases. This ELISA works well for low level detection of TNF in mouse serum and from macrophage cell cultures. This was easy to use and set up with a standard curve to allow for absolute pg/mL amounts to be determined. The kit works reliably each time.

Protocol Overview

Mouse serum was collected from LPS challenged mice and surveyed in this ELISA. Serum was places in the ELISA overnight at 4C and washed out as per protocol. Secondary and HRP was added subsequently as per protocol.

Tips

Titrate serum dilutions, I used 1:10 for optimal range detection.

Results Summary

Works well, may need to optimize with dilutions of samples depending on sample amounts and concentration of TNF.
