I used this antibody detect CD4+IFN-g+ Th1 cells by flow cytometry. Total splenocytes were stimulated with PMA ionomycin + brefeldin A for 4 hr and cells were fixed/permeabilized, and stained with CD4 and IFN-g antibody for flow cytometry analysis.
