Fluorescent phalloidin was used to stain neuronal dendrites for spine density counts in pre-adolescent compared to adolescent mice. At the scale of spines the dilution required to visualize and do counts using confocal microscopy was 1:20. Scale bar of image is 10 micrometers.

Results Summary

To detect and do neuronal spine density counts, a dilution of 1:20 was required of this antibody. Additionally, the resolution of images was highly dependent on the quality of the perfusion and only with overlapping stains was there accurate consensus of whether the stain was staining the appropriate structures in certain cases. Scale bar of images is 2.5 micrometers.

The Bottom Line

The idea of an antibody conjugated with fluorescence is appealing; however, this antibody required a very high concentration to produce usable stained mouse brain slices.
