Immunofluorescence
Human Spleen (FFPE) / Human primary macrophages in 3D.
1:200 (in blocking buffer), overnight at 4°C (1:100 for macrophages in 3D)
TBS 10% goat serum, 1% BSA (1hr @ RT)
1:1000, AlexaFluor 488 in TBS 1% BSA, 1hr @ RT (1:500 for macrophages in 3D)

Strong and specific signal. Figure 1. Spleen tissue. CD68 can be seen in the red pulp of the spleen, where macrophages (CD68 is a macrophage marker) can be found. Figure 2. CD68 (green) and CD163 (red) in human primary macrophages were derived from peripheral blood monocytes from healthy donors, and encapsulated in 3D hydrogel composed of hyaluronic acid and collagen type 1. Cells were then fixed in 4%PFA for 10 minutes, permeabilized with TBS 0.25% triton X during 3X5min washes; blocked with blocking buffer and incubated as described above.
