TLR2 and TLR4 signaling plays a pivotal role in macrophage activation. Therefore, we used Pam2CSK4, a TLR2 agonist, and LPS, a TLR4 agonist, to stimulate macrophages.

Raw macrophages were stimulated either with LPS (200 ng/ml) or Pam2CSK4 (50 nM) for 8 h to follow TNFa concentration in cell culture medium using ELISA.
