The aim of this experiment was to digest whole rat lung using the Liberase™ TL Research Grade reagent. The enzyme worked well and the protocol was easy to implement. The reagent was recommended by colleagues who had used it for harvesting single cells from mouse lung.
Digestion of tissue
Whole lung tissue (sections)
Digestion with Liberase™ TL Research Grade for 10 min and then wash with buffer comprising of RPMI (no phenol) and 5% fetal bovine serum to inactivate the enzyme.
Perform reaction at room temperature.
Single cells were isolated from the rat lung tissue.
