Western blots have been one of the most common techniques for quantifying protein expression in a biological sample. Although most WB procedures take 1–2 days to perform, nowadays companies provide reagents that can speed up the process and provide more consistent results. Here, I review the MIDI Bio-Rad Trans-Blot pack for transferring up to two gels onto PVDF. It is convenient and easy to use.

Experimental Design and Results Summary

Application

WB

Starting Material

A375 whole-cell lysate

Protocol Overview

The pack comes with pre-stacked pads with a PVDF membrane in the middle. All the user needs to do is to place the pad and the PVDF onto the transfer apparatus and then add the gel. The top pad can be added right on top of the PVDF to complete the gel-PVDF sandwich. A roller is used to remove any bubbles.

Tips

Try to maximize the transfer by using this MIDI pack for 2 gels

Results Summary

A Turbo transfer can be used to complete the transfer of two gels in ~7 mins. A standard transfer can also be done using 2 gels in ~30 mins. Can also transfer one gel if needed.
