We used this kit in experiments in which we were investigating Fc-mediated functions of NK cells in the context of HIV infection. For this we needed to isolate NK cells from human PBMCs. This kit was used and consistently resulted in a high-purity yield of human NK cells.

Protocol Overview

Isolate human PBMCs from whole blood. Wash, count cells, incubate with the biotin-labeled antibody cocktail followed by labelling with beads. According to manufacturer's instructions, apply labeled cells to column, wash 3X, elute. In the second step, label the target cells with CD16 beads and repeat the separation process with column.

Tips

Titrate the beads to save some beads, usually the amount of beads/antibody can be decreased while still giving the same isolation efficiency

Results Summary

The recovered cells show high purity of CD56-expressing and CD16-expressing NK cells.
