I am interested in studying the molecular mechanism of hedgehog signal transduction in vertebrate cells. I routinely perform western blotting for target genes and core hedgehog pathway components to measure the signaling output and monitor baseline levels, respectively. Orbital shakers form an integral part of the western blotting process in which blotting membranes are washed and incubated with antibodies. We had purchased the Helix 150 Orbital Shaker based on a recommendation from a neighboring lab.
Western Blotting
Western Blotting: membranes (placed in a small box) are incubated with washing buffer (1x TBS-T) or antibody solutions and subjected to gentle shaking either for a few minutes or for 1–2 hours at room temperature.
