As an immunology lab, we routinely analyze the ability of T cells to make IL-2 since it's the first cytokine T cells make. The amount a T cell makes is very important since this cytokine helps T cells grow in an autocrine fashion at first. We stimulate T cells and assess IL-2 secretion using flow cytometry.

We are able to detect IL-2 expression using flow cytometry in CD8 T cells upon stimulation. We identified cells that make robust to moderate amounts of IL-2 but some that make none.
