As an immunology lab, we perform various molecular biology–based experiments where we routinely do PCR or western blot analysis. This imaging system allows us to easily view DNA gels and also use the instrument for chemiluminescence for immunoblotting analysis. The acquisition of the gels and blots using a computer makes it all digital rather than using film.

Protocol Overview

Depending on whether it's being used for DNA or western blot adjust the stage. Select the correct preset settings within the application to adjust the lighting (UV for DNA and Chemi for western) and camera settings. With the camera setting, determine the length of the exposure. For DNA gels, we typically expose for 1–2 seconds. For western blotting, we do a minimum of 1 minute to a maximum of 60 minutes (for weak signals).

Tips

All depends on the reagents used for DNA. Western blotting is highly dependent on the reagents such as antibodies including secondary HRP and ECL substrate.
