Buffering the media is very critical to maintain normal pH levels for cultures. HEPES is an additional buffering system we use in conjunction with the standard carbonate buffering system.
Buffering Media
T cell media with all components
Make up the final media with all the components such as antibiotics, RPMI, and serum for 1 L. Add 10 mL of the 100x HEPES solution.

Buffering with HEPES keeps the media fresher for longer periods, and the cells are much happier and healthier.
