I'm interested in understanding the molecular mechanism of hedgehog signal transduction. Some of the experiments used for measuring the pathway activity include luciferase assays and protein estimations (BCA or Bradford). This product was extensively tested during demo phase and then purchased by our lab with the dual injection port module for luciferase assays.

Protocol Overview

Cells were transfected with the Firefly luciferase and Renilla luciferase constructs. After specific treatments, cell lysates were prepared and the Firefly and Renilla luciferase activities were measured in this system by using the dual injection port module (well-by-well).

We had purchased both monochromator-based optics and filter-based optics for flexibility and sensitivity. We also opted for top and bottom fluorescence intensity measurement. This is one of our most widely used instruments in the lab and has had no issues for the past 4 years.
