I'm interested in understanding the molecular mechanism of hedgehog signal transduction. The experimental model system that I use is a mouse fibroblast cell line. I routinely perform western blotting for the pathway target genes and this platform shaker is used during the primary and secondary antibody incubation steps of the blotting membranes. I also use during the wash steps.
Western Blotting
Western Blotting: SDS-PAGE resolved proteins are transferred onto nitrocellulose membranes. Membranes are incubated with primary antibodies (overnight at 4°C). Membranes are washed thrice (10 min each wash step), followed by secondary antibody incubation (1 hour at room temperature). Membranes are washed again and then proceeded for chemiluminescence detection.
No specific steps. Straightforward to use.
I prefer the double platform version because we are a lab of extensive western blotting users.
