We routinely use TRIzol to extract RNA from tissue or single-cell suspended cells after pelleting. This solution does everything since it lyses the cells while freeing the RNA. The procedure to extract RNA is quite straightforward also.

Protocol Overview

Pellet cells then remove supernatant. Resuspend the pellet in appropriate volume of TRIzol using filtered tips. Let lyse for 10 minutes at room temp before storing in –80°C or proceeding with RNA extraction using chloroform, isopropanol, and ethanol, depending on volume used.

Tips

Determine optimal volume of TRIzol to use based on cell number and cellular content.

Results Summary

We are able to recover high yield and clean RNA following the protocol.
