Our lab studies the role of mTOR signaling in CD8 T cell differentiation and function. We routinely conduct experiments with splenocytes that need removal of red blood cells (RBCs) before we can proceed with downstream applications. We use this buffer for that exact purpose of removing the RBCs.

Protocol Overview

Excise and process spleen to single-cell suspension. Pellet suspension then remove the sup. Use 2–3 mL of ACK buffer for about 2 minutes until RBCs are no longer present. Add 30 mL of PBS to bring back to isotonic concentration.

Tips

Make sure you bring back the tonicity with PBS.

Results Summary

We are able to remove all RBCs with simple ACK lysing.
