Our goal was to examine the global levels of H3 in lysates of HeLa and 293T cells by western blot. We lysed both the cell lines in NETN buffer, sonicated the samples and loaded equal amount of cell lysates on SDS-PAGE and transferred on to PVDF membrane. H3 levels were analysed by Western blot using H3 specific antibody.
