Immunofluorescences were done in our laboratory to study the effects of toxic molecules on organotypic brain cultures. We stained neurons and astrocytes with different secondary antibodies and counterstained with DAPI.We selected the product because we need a mounting media that allow a strong intensity of the signal, a clear image and stability of the brain section for a long time.

Protocol Overview

The mounting media were used at the end of the process before the cover-glass. If the staining of the nucleus is low, we incubated 5 minutes with DAPI 1:2000.

Tips

Check if the DAPI present in the media is enough for you application; on ther contrary add a short incubation with DAPI.

Results Summary

We obtained clear images, with a well-stained nucleus. The images were stable for months with a strong fluorescence.
