Company:

Invitrogen

Product Name:

CD235a Monoclonal Antibody (CLB-ery-1 (AME-1)), FITC

Catalog Number:

MHGLA01

The aim of our research is to detect paroxysmal nocturnal haemoglobinuria (PNH) on red blood cells (RBC). We needed a good antibody that stains the RBCs to distinguish them from debris and other small cells in peripheral blood. We chose this antibody because it was recommended to us by the clinical lab at UK.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Whole blood

Primary Incubation

5uL antibody [15 minutes at room temp (in the dark)]

Blocking Agent

None

Secondary Incubation

Detection

Detected on the FITC (FL-1) channel BP530/30

Results Summary

The results show very good staining of the RBCs with the FITC-conjugated antibody. As shown in the image, we used FSC/SSC in log to gate on the RBCs (gate: P1) and then check their staining with CD235a using a histogram. Positive cells are shown in gate P2.

Summary

The Good

Strong staining

The Bad

May take a while to titrate it to the optimal concentration

The Bottom Line

A good antibody, which identifies RBCs clearly. The fact that it's ASR is an added bonus!
