Company:

Santa Cruz Biotechnology

Product Name:

OctA-Probe Antibody (H-5)

Catalog Number:

sc-166355

My experiment was focused on evaluating the effect of a variant on the localisation of a cytoplasmic protein. I overexpressed wild-type and mutant isoforms of the protein tagged with FLAG and studied their localisation. The OctA-Probe antibody was used for immunofluorescence on HEK293 cells transiently transfected with a FLAG-fused protein.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

HEK293 cell line

Primary Incubation

Antibody diluted 1:100 in blocking solution, incubated for 2 hrs at RT

Blocking Agent

BSA 2%

Secondary Incubation

goat anti-mouse AlexaFluor 555 (A28180) diluted 1:200 in blocking solution 1 hr at RT

Tertiary Incubation

Detection

Olympus FluoView FV1000

Results Summary

The antibody performs well in immunofluorescence on paraformaldehyde fixed cells. Low background as compared to stain performed on non transfected cells. It also works with a 1:200 dilution.

Summary

The Good

Specific stain and bright signal also at higher dilutions

The Bad

Nothing at the moment

The Bottom Line

I only tested the antibody in immunofluorescence and it is specific and easy to optimise.
