Phosphorylation of S6 protein is used as a marker to study the activation or inhibition of mTORC1 complex in multiple studies.Our goal was to examine the activity of mTORC1 signaling in HeLa cells by checking the levels of phospho-S6 (ser240/244) levels in cell lysates. We have transfected HeLa cells with a Scramble and mTOR siRNA for 48 hrs. Whole cell lysates were prepared after transfection and the activity of the mTORC1 complex was analyzed by western blot using phospo-S6 (ser240/244) antibody.
