Product Name:

Phospho-S6 Ribosomal Protein (Ser240/244)

Catalog Number:

2215S

Phosphorylation of S6 protein is used as a marker to study the activation or inhibition of mTORC1 complex in multiple studies. Our goal was to examine the activity of mTORC1 signaling in HeLa cells by checking the levels of phospho-S6 (ser240/244) levels in cell lysates. We have transfected HeLa cells with a Scramble and mTOR siRNA for 48 hrs. Whole cell lysates were prepared after transfection and the activity of the mTORC1 complex was analyzed by western blot using phospo-S6 (ser240/244) antibody.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Control and mTOR siRNA treated HeLa whole cell lysates

Primary Incubation

1:5000, Overnight at 4'c

Blocking Agent

5% skimmed milk, 1 hr at room temperature

Secondary Incubation

1:10000, anti-rabbit HRP conjugated antibody, 1hr at room temperature

Tertiary Incubation

Results Summary

This antibody detects a specific phospho-S6 band at the 32KD molecular weight on SDS-PAGE. We observed a decreased phosphorylation of S6 in mTOR knockdown lysates indicating the specificity of antibody used
